show Abstracthide AbstractWe have isolated 66 strains of bacteria and 2 strains of archaea from a Japanese centenarian using Brucella agar plate with haemin, Vitamin K1 (BHK) agar, BHK agar containing paramomycin and vancomycin, and reinforced clostridial (RC) agar under anaerobic conditions. Individual colonies emerged after 72 hr and up to 10 days of incubation were picked. Isolated strains were identified by PCR amplification of the 16S rRNA gene region with universal primers. The extracted genomic DNA of 68 isolated strains was sequenced using PacBio Sequel and Illumina MiSeq sequencers.
